DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Several antiinflammatory steroids and their derivs., but not dexamethasone (I) [50-02-2] or I monosodium phosphate [1869-92-7], at concns. of 10-3-10-7M, decreased collagen formation in chick embryo tendon cells. The amt. of labeled collagen decreased more in the medium than in the cells, but no steroid caused an accumulation of labeled collagen in the cells. Hydrocortisone (II) [50-23-7] was as inhibitory as hydrocortisone-17-butyrate [13609-67-1], but both were more effective than II Na succinate [125-04-2] or II monosodium phosphate [38048-30-5]. Betamethasone-17-valerate [2152-44-5] was the most potent inhibitor tested, but betamethasone (III) [378-44-9] and III disodium phosphate [151-73-5] were ineffective. Prednisolone [50-24-8] was as effective against collagen as II. The decrease in collagen followed the decrease in total labeled protein. Inhibition of collagen formation by antiinflammatory steroids may be a consequence of a general protein formation inhibition.
