DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Sensitive gas chromatog. procedures for the detn. of methoxyphenamine (I) [93-30-1] and 3 of its metabolites in plasma have been developed. The metabolites were measured with an electron-capture detector. This simple procedure is based on the pptn. of protein from a 1-mL plasma sample with 10% Cl3CCO2H, followed by derivatization with pentafluorobenzoyl chloride at pH 9.2 and a single-step cyclohexane extn. The lower limit of detection for the N-demethyl [15402-84-3], O-demethyl [582-43-4] and arom. 5-hydroxy [61866-76-0] metabolites of methoxyphenamine were 1.6, 3.1 and 2.2 ng/mL, resp., with coeffs. of variation <10%. The poor electron-capture response of fluorinated derivs. of methoxyphenamine necessitated the use of N-P detection. Extractive derivatization with pentafluorobenzoyl chloride, without the need for protein pptn., enabled quantitation of methoxyphenamine at 33.8 ng/mL from a 2-mL aliquot of plasma. In a pilot study involving healthy volunteers who received a single oral dose of methoxyphenamine hydrochloride, plasma concn. could be followed in all 3 subjects for at least 24, 32, 12 and 4 h for methoxyphenamine and the O-demethyl, 5-hydroxy and N-demethyl metabolites, resp.
