DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 28 cis-Mafosfamide (I) [84210-72-0], a stable analog of cis-4-hydroxycyclophosphamide (II) [61903-29-5], undergoes rapid decompn. in aq. phosphate buffer or plasma at pH 7.4 and 37°. The reaction kinetics of I complex, and trans-mafosfamide (III) [92745-18-1] and II are produced and subsequently disappear over the course of the reaction. The rates of decompn. of I as well as III are much faster in plasma than in buffer. The cis-trans isomerization of I occurs by a specific-base-catalyzed process via iminocyclophosphamide [84489-09-8] as a transient intermediate. In contrast, formation of cis- and trans-mafosmafmide from III and Mesna [19767-45-4] proceeds by an acid-catalyzed process via a hemithioacetal intermediate. The significance of these findings with respect to cyclophosphamide [35144-64-0] metab. is discussed. .
