DOCUMENT TYPE: Journal CA Section: 3 (Biochemical Interactions) Section cross-reference(s): 1 The administration of 4 mg/kg/day of dichloromethane diphosphonate disodium salt (I) [22560-50-5] s.c. to pregnant cows fed a low-Ca diet significantly decreased bone resorption. Plasma parathyroid hormone [9002-64-6] levels were similar between I-treated and control cows prepartum, during EDTA infusions, and near parturition. Ultrastructurally, chief cells of the parathyroid glands of both groups of cows were in the active stage of the secretory cycle. The chronically stimulated chief cell from cows administered I had a large cytoplasmic area contg. many lipofuscin granules and lysosomal bodies and a few secretory granules near the large Golgi app. or aligned along the plasma membrane. Uptake of Ca by the duodenal mucosa incubated in vitro was greater in I-treated cows compared to control cows. The administration of I significantly decreased rapidly mobilizable Ca reserves. Following an i.v. EDTA infusion and the spontaneous Ca drain assocd. with parturition and the beginning of lactation, I-treated cows developed severe hypocalcemia. The rapid mobilization of Ca reserves in cows administered I prepartum was impaired mainly because of diminished resorption of bone despite adequate parathyroid hormone secretion in response to severe postpartal or EDTA-induced hypocalcemia.

DOCUMENT TYPE: Journal CA Section: 3 (Biochemical Interactions) Inorg. pyrophosphate and disodium dichloromethane diphosphonate [22560-50-5] had essentially no effect on the ATP-driven transport of Ca from sealed human erythrocyte ghosts. This was true whether the compds. were introduced into the cells or added to the external medium. Disodium ethane-1-hydroxy-1,1-diphosphonate [7414-83-7] slightly decreased the rate const. for Ca transport when within the ghosts, but increased it slightly when present in the external medium. Thus, neither the pyrophosphate nor the 2 diphosphonates had any major effect on Ca transport by erythrocytes. Incubation of ghosts contg. labeled compds. resulted in little loss of radioactivity relative to the leaking of Hb, indicating that the erythrocyte membrane was relatively impermeable to pyrophosphate and the tested diphosphonates.
