DOCUMENT TYPE: Journal CA Section: 15 (Immunochemistry) The effects of polyinosinic, polycytidylic acid [poly(I)×poly(C)] on the activation and RNA metab. in murine peritoneal macrophages (MF) elicited by proteose-peptone (pMF) was investigated. 24939-03-5 is also in the experiment. Poly(I)×poly(C) triggered the cytolytic activity of pMF and augmented their glucose oxidn. In contrast, a profound depression of [3H]uridine incorporation into RNA was obsd. in poly(I)×poly(C)-activated pMF. The degree of depression of RNA labeling paralleled the dose of poly(I)×poly(C) used to activate the pMF and the expression of tumoricidal activity. This decrease in [3H]uridine incorporation into MF RNA could not be accounted for by decreased permeability of the activated MF to [3H]uridine, or by instability of the labeled RNA. The decreased RNA labeling was not due to changes in the specific activity of UTP. Thus, poly(I)×poly(C)-activated pMF exhibit a depressed rate of RNA synthesis. The rate of RNA synthesis may be a new indicator for MF activation. .

DOCUMENT TYPE: Journal CA Section: 15 (Immunochemistry) Purified interleukin 2 (IL-2), free of interferon (IFN), enhanced natural killer (NK) activity of normal human peripheral blood mononuclear cells (PBMC). This enhancing activity was absorbed by IL-2 receptor-bearing cells, but was not blocked by antibody to a-IFN. IFN in the culture supernatants was greatly increased after stimulation with poly(I:C) plus IL-2. There was less IFN produced by either modulator acting alone. 24939-03-5 which is the cas registry number of some chemical is mentioned. Stimulation of PBMC with IL-2 and/or poly(I:C) increased the proportion of OKM1+ cells and anti-Leu-7+ cells. When cells expressing either surface antigen were specifically lysed to deplete NK, cytotoxic activity could be restored by overnight incubation in IL-2. This suggests that IL-2 stimulates the development of NK cells from precursors that lack cell surface OKM1 or Leu-7 antigens. IL-2 acted directly on large granular lymphocytes and did not require the presence of adherent cells. Probably, IL-2 acts synergistically with other IFN inducers and may play an important role in the regulation of NK cells. .
