A very simple sample prepn. consisting of an extn. with an org. solvent was carried out. Sample exts. were injected into the LC/MS/MS system on a C18 column and an isocratic elution was performed. Nigericin was used as internal std. The precursor ions produced by electrospray pos. ionization were selected for collisional dissocn. with argon into product ions. Validation of the methods was performed based on Commission Decision 2002/657/EC. CCa was found to be 1 mg/kg for all four compds. Monitoring of Belgian egg samples in 2004 revealed that residues of salinomycin, lasalocid and monensin could be found.

DOCUMENT TYPE: Journal CA Section: 10 (Microbial, Algal, and Fungal Biochemistry) A set of expts. was carried out to test hypotheses on the relation between the microtubule (MT) cytoskeleton and tip growth in fungal hyphae. Using a growth assay with 1.5 h-old germ tubes of P. infestans, various chems. that inhibit growth or disrupt MTs or that affect both were tested. Germ tubes were measured after 2 h exposure to the test compds. and MTs were revealed by immunofluorescence. The phytoalexin glyceollin had a dual effect, as it stimulated germ tube growth at concns. of 10-5M and 10-6M, but inhibited it at 10-4M. It affected the MT cytoskeleton at neither of these concns. Metalaxyl, the active ingredient of the fungicide Ridomil, inhibited germ tube growth slightly at 10-4M and 10-6M, but it had no effect on MTs. Benomyl, the active ingredient of the fungicide Benlate, had no effect on MTs at a concn. of 10-5M, although it partially inhibited germ tube growth. Nocodazole, a benzimidazole inhibitor of MTs, disrupted the subapical MT cytoskeleton in germ tubes of P. infestans at 10-4M, at which growth was still 79% of the control. The K+ ionophore nigericin and the Ca2+ ionophore A23187 stimulated germ tube growth at concns. between 10-6M and 10-9M, but inhibited it at 10-4M. Nigericin, but not A23187, disrupted MTs at this concn. These results point out that no simple relation exists between an intact MT cytoskeleton and germ tube growth in P. infestans.
