DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Geodin (I) [427-63-4], trypacidin (II) [1900-29-4], plumbagin (III) [481-42-5], terreic acid [121-40-4] and monorden [12772-57-5] were potent inhibitors of glyceraldehyde-3-phosphate dehydrogenase (EC 1.2.1.12) [9001-50-7] and other sulfhydryl (SH) enzymes, glucose-6-phosphate dehydrogenase (EC 1.1.1.49) [9001-40-5], glutathione reductase (EC 1.6.4.2) [9001-48-3] and malate dehydrogenase (EC 1.1.1.37) [9001-64-3]. The expts. with reduced glutathione [70-18-8], as the model low mol. thiol, showed that these compds. were reactive thiol-combining agents. The ability to react with SH groups is discussed in the light of the biol. activities of these naturally occurring a,b-unsatd. carbonyl compds. Structure-activity relations are discussed.
