DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) Section cross-reference(s): 11, 63 Gentianine (I) [439-89-4] and gentianal (II) [53848-05-8] were not detected by TLC in the EtOH ext.Several reagents with their cas registry numbers 11019-08-2 and 439-89-4 are used here. of Qin Jiu (dried root of Gentiana species, including G. macrophylla, G. stramina, G. crassicaulis, and G. dahurica); however, I and II were detd. when the EtOH ext. was mixed with a NH4OH soln. and allowed to stand for 24 h. Similarly, no I and II were detected when Qin Jiu was extd. with Na2CO3-CHCl3 or MeOH-HCl-Na2CO3-CHCl3, whereas I and II were detected when Qin Jiu was extd. with NH4OH-CHCl3 and analyzed by TLC on silica gel GF254 using CHCl3-EtOH () as developing solvent system. In addn., I and II were produced on treatment of o-tetraacetylgentiopicroside [11019-08-2] extd. from Qin Jiu with NH4OH, indicating that I and II detected in Qin Jiu are not natural constituents, rather they were byproducts of NH3 treatment. Thus, the recommended method in the Chinese Pharmacopia, which used I and II as marker constituents for identification of Qin Jiu, may be questionable. .

Results from m.p. detn. and mass, NMR, IR, and UV spectroscopy indicated that 4 of the constituents were gentianine, gentianidine, gentianal, and b-sitosterol. .
