DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The biol. transformation of phenyramidol (I) [553-69-5], some of which was also excreted unchanged, occurred by 3 main degradative pathways: hydroxylation of the pyridine ring in position 3 and 5, cleavage of the ethanolamine chain with the formation of 2-aminopyridine [504-29-0] and presumably mandelic aldehyde, and conjugaton with glucuronic acid. Secondary reactions resulted in the prodn. of benzoyl carbinol [582-24-1], benzoic acid [65-85-0], mandelic acid [90-64-2], and the glucuronides of several metabolites which were also found as free, unconjugated compds. A further, unusual reaction is the dimerization of a hydroxy metabolite of I with the formation of a dipyridyl deriv., which is excreted partly as the free compd., but mainly as the glucuronide. The occurrence of 2-(N-benzylamino)pyridine [6935-27-9] in the urine could not be explained. Four further excretory products were not identified; 2 of them were extd. at an alk. 553-69-5 is also in the experiment. pH, whereas the other 2 were extd. under neutral conditions. They could be detected both as free compds., and after hydrolysis with HCl or alkali, but not after treatment with b-glucuronidase. ..
