DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The uterotonic activity of vasicine (I) [6159-55-3] was investigated in detail on the uteri of different species of animals and in different hormonal states both in vitro and in vivo. The uterotonic activity was similar to that of oxytocin and methylergometrine. The uterotonic effect of I was influenced by the degree of priming of the uterus by estrogens (known to enhance the synthesis of prostaglandins in the uterus) and it was markedly decreased after pretreatment of the uterus with aspirin and indomethacin. Thus, the uterotonic effect of I is at least partly mediated through the release of prostaglandins.

Vasicinone (II) [486-64-6] showed bronchodilatory activity in vitro and bronchoconstriction in vivo. The 2 alkaloids in combination had more bronchodilatory activity in vitro and in vivo. I also had marked respiratory and uterine stimulant activity and moderate hypotensive activity. II was devoid of these activities.
