DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effects of sodium diethyldithiocarbamate (DDTC) [148-18-5], sodium thiosulfate (TS) [7772-98-7] and D-penicillamine (PN) [52-67-5] on the nephrotoxicity and antitumor activity of the antitumor agent, cisplatin [15663-27-1] were investigated in mice. The acute toxicity of cisplatin (20 mg/kg) was prevented by the treatment with DDTC (1,000 mg/kg) 45 min after or TS (1,500 mg/kg) 30 min before the treatment with cisplatin. The administration of PN (800 mg/kg) 30 min before the treatment with cisplatin reduced the toxicity of cisplatin. DDTC and TS protected the body and kidneys from the wt.-loss induced by cisplatin and reduced the increase induced by cisplatin in the value of blood urea N. Cisplatin (20 mg/kg) had no effect on the formation of lipid peroxide in kidneys. The administration of DDTC, TS, and PN decreased the antitumor activity of cisplatin (10 mg/kg) against the solid form of Ehrlich tumor.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) With the use of phytohemagglutinin (PHA) stimulation assay of human peripheral blood mononuclear cells (PBM), the protective effect of Na thiosulfate (STS) [7772-98-7] on the antiproliferative action of cis-diamminedichloroplatinum(II) (DDP) [15663-27-1] against human cell was investigated. DDP alone inhibited the proliferation of PBM over the concn. range of 10-7 to 5 ′ 10-5 M. The antiproliferative effect of DDP was blocked when STS was added to the culture at the time of exposure to DDP, at molar STS/DDP ratios of >500. However, STS at molar ratios of <100 induced minimal protection. Protection from 1000-fold excess of STS was effective within 10 min after exposure to DDP at a concn. of 5 ′ 10-5 M, whereas it was not effective beyond 30 min after the exposure.
