Synthesis of 2-amino-1-(4′-methoxyphenyl)-propane (II) starting from p-anisaldehyde (1) in 67% overall yield is described. Key reactions involved Horner-Wadswoth-Emmons olefination of 1 to form the ester 2 and Hoffmann degradation of amide 5 to obtain the amine II.

In this Communication, procedures for the selective synthesis of 4-arylbutanoic acids, 2-arylbutanoic acids, and 4-arylbutanals from the same allylbenzenes have been developed. With formic acid or TFBen as the CO surrogate, reactions proceed selectively and effectively under carbon monoxide gas-free conditions.

Methods of making such therapeutic and/or protective compounds and associated compositions are also provided, as are methods for their use, which include protecting cells from oxidative damage and/or inhibiting production of ROS in a cell or subject, as well as preventing or reducing the extent of tissue damage caused by an ischemic event in a subject at elevated risk of such an event.
