- Saccharide-modified nanodiamond conjugates for the efficient detection and removal of pathogenic bacteria

The detection and removal of bacteria, such as E. coli in aqueous environments by using safe and readily available means is of high importance. Here we report on the synthesis of nanodiamonds (ND) covalently modified with specific carbohydrates (glyco-ND) for the precipitation of type 1 fimbriated uropathogenic E. coli in solution by mechanically stable agglutination. The surface of the diamond nanoparticles was modified by using a Diels-Alder reaction followed by the covalent grafting of the respective glycosides. The resulting glyco-ND samples are fully dispersible in aqueous media and show a surface loading of typically 0.1 mmol g-1. To probe the adhesive properties of various ND samples we have developed a new sandwich assay employing layers of two bacterial strains in an array format. Agglutination experiments in solution were used to distinguish unspecific interactions of glyco-ND with bacteria from specific ones. Two types of precipitates in solution were observed and characterized in detail by light and electron microscopy. Only by specific interactions mechanically stable agglutinates were formed. Bacteria could be removed from water by filtration of these stable agglutinates through 10 μm pore-size filters and the ND conjugate could eventually be recovered by addition of the appropriate carbohydrate. The application of glycosylated ND allows versatile and facile detection of bacteria and their efficient removal by using an environmentally and biomedically benign material. Copyright

Phosphomolybdic acid supported on silica gel provides a truly catalytic method for the chemoselective cleavage of benzylidene acetals having sensitive functional groups under mild conditions. It is easy to perform on large scale owing to minimal catalyst loading (0.5 mol-%). Several sensitive functional groups such as TBDPS ether, -OMs, -OAc, allyl ether, N-Boc, N-Fmoc and N-Cbz are stable under the reaction conditions. - Synthesis and study of the behavior of glycosylated gemini surfactants in aqueous media

The work reported herein deals with the synthesis and physico-chemical studies of a new kind of glycosylated non-ionic gemini surfactants. These compounds derive from tris(hydroxymethyl)aminomethane and bear hydro- or perfluorocarbon tails. The chemical structures of the spacer arm and hydrophobic tails were adjusted in order to specify their impact on the gemini surfactant behavior in aqueous media. The supramolecular systems they form were studied by TEM after negative staining. The length and the rigidity of the spacer arm seem to play a key role in the aggregation behavior of these surfactants in water. With the appropriate modifications, the formation of premicellar associations, micelles, vesicles or various aggregates can be observed.

- A Convergent Synthesis of Carbohydrate-Containing Dendimers

The synthesis of carbohydrate-containing dendrimers has been achieved by a convergent grwoth approach.The synthetic strategy involves: 1) the synthesis of the triglucosylated derivative of tris(hydroxymethyl)methylamine (TRIS), 2) the introduction of a glycine-derived spacer and 3,3'-iminodipropionic acid derived branching units on to the TRIS derivative by amide bond formation, 3) condensation of the above saccharide-containing dendrons with a trifunctional 1,3,5-benzenetricarbonyl derivative, used as the core, by formation of amide bonds, and 4) deprotection of the saccharide units.A 9-mer and an 18-mer, carrying nine and eighteen saccharide units at the periphery, respectively, have been synthesized, in high yields at each step, by this synthetic strategy.By a variety of chromatographic and spectroscopic techniques, the dendrimers were shown to be structurally homogeneous, monodisperse, and error-free at all steps in their growth.These investigations were complemented by molecular modeling studies on the dendrimers.The presence of slightly distorted C3 symmetry was noted in both the 9-mer and the 18-mer. - Keywords: carbohydrates; cluster glucosides; convergent syntheses; dendrimers; neoglycoconjugates
