RAS-2410 hydrochloride is an orally bioavailable inhibitor of histone deacetylase 1 (HDAC1), HDAC3, and HDAC6, with IC50s ranging from 43-72 nM. It leads to hyperacetylation of histone H4 in multiple myeloma cells, effectively abrogating cell growth and inducing apoptosis (IC50s = 2.5-3 μM). RAS-2410 hydrochloride also exhibits synergistic effects when combined with melphalan, bortezomib, and S-2209, and it dose-dependently inhibits HDAC activity in vivo.
