Two-photon imaging, due to its unique advantage to utilize near-infrared light as the excitation source, has emerged as an important tool for biomedical research. Herein, the first NBD amine based two-photon fluorescence probe L using a FRET strategy was developed for detecting H2S in vitro and in vivo. The probe L not only afforded high selectivity and sensitivity for the detection of H2S, but also displayed a linear response to H2S with a low detection limit of 24 nM. Furthermore, the living cell experiments indicated that L can be effective for the both exogenous and endogenous detection of H2S inside living cells by two-photon fluorescence imaging.

The invention provides a 7-nitro-1,2,3-benzoxadiazole (NBD) derivative and a synthesis method and application thereof, and belongs to the field of detection of NBD derivatives and mercury ions. The synthesis method of the NBD derivatives comprises the following steps of: first, dissolving 4-chlorine-7-nitrobenzo-1,2,3-benzoxadiazole and anhydrous piperazine in 2-methoxyethanol, heating and refluxing, and separating by column chromatography to obtain a compound 1; then, heating and refluxing 2,6-bis (chloromethyl) pyridine and 8-hydroxyquinoline in an acetone solution, and separating by the column chromatography to obtain a compound 2; finally, dissolving the compound 1 and the compound 2 in acetonitrile, refluxing for 12 hours, and separating by the column chromatography to obtain the product NBD derivative. The invention also provides a method for detecting the mercury ions, which comprises the step of quantitatively detecting the content of Hg in a solution of all water (pH = 7.4) by using the NBD derivative as a probe. The detection method can detect the mercury ions in an aqueous solution with high selectivity and sensitivity, and is simple, convenient and rapid to operate.Meanwhile, test paper for detecting the mercury ions is provided, so that the Hg detection is more convenient.

- Dual-Reactable Fluorescent Probes for Highly Selective and Sensitive Detection of Biological H2S
-
Hydrogen sulfide (H2S) is an important endogenous signaling molecule with a variety of biological functions. Development of fluorescent probes for highly selective and sensitive detection of H2S is necessary. We show here that dual-reactable fluorescent H2S probes could react with higher selectivity than single-reactable probes. One of the dual-reactable probes gives more than 4000-fold turn-on response when reacting with H2S, the largest response among fluorescent H2S probes reported thus far. In addition, the probe could be used for high-throughput enzymatic assays and for the detection of Cys-induced H2S in cells and in zebrafish. These dual-reactable probes hold potential for highly selective and sensitive detection of H2S in biological systems. Two heads are better than one: Fluorescent probes with two types of reactive heads (R1 and R2) for H2S can lead to a higher selectivity than that of single-reactable probes (See Figure). The probe was used for highly selective and sensitive detection of biological H2S.

Hydrogen sulfide (H2S) is considered as the third signaling molecule in vivo and it plays an important role in various physiological processes and pathological processes in vivo, such as vasodilation, apoptosis, neurotransmission, ischemia/reperfusion-induced injury, insulin secretion and inflammation. Developing a highly selective and sensitive method that can detect H2S in the biological system is very important. In this work, a colorimetric and "turn-on" fluorescent probe is developed. Furthermore, this probe displays a highly selective response to H2S in aqueous solution and possesses good capability for bioimaging H2S without interference in living cells. The results suggest that a H2S-selective probe has good water-solubility, biocompatibility and cell-penetrability and can serve as an efficient tool for probing H2S in the cell level.
