A novel two-photon fluorescent probe for hydrogen sulfide in living cells using an acedan-NBD amine dyad based on FRET process with high selectivity and sensitivity

Two-photon imaging, due to its unique advantage to utilize near-infrared light as the excitation source, has emerged as an important tool for biomedical research. Herein, the first NBD amine based two-photon fluorescence probe L using a FRET strategy was developed for detecting H2S in vitro and in vivo. The probe L not only afforded high selectivity and sensitivity for the detection of H2S, but also displayed a linear response to H2S with a low detection limit of 24 nM. Furthermore, the living cell experiments indicated that L can be effective for the both exogenous and endogenous detection of H2S inside living cells by two-photon fluorescence imaging.

The invention provides a 7-nitro-1,2,3-benzoxadiazole (NBD) derivative and a synthesis method and application thereof, and belongs to the field of detection of NBD derivatives and mercury ions. The synthesis method of the NBD derivatives comprises the following steps of: first, dissolving 4-chlorine-7-nitrobenzo-1,2,3-benzoxadiazole and anhydrous piperazine in 2-methoxyethanol, heating and refluxing, and separating by column chromatography to obtain a compound 1; then, heating and refluxing 2,6-bis (chloromethyl) pyridine and 8-hydroxyquinoline in an acetone solution, and separating by the column chromatography to obtain a compound 2; finally, dissolving the compound 1 and the compound 2 in acetonitrile, refluxing for 12 hours, and separating by the column chromatography to obtain the product NBD derivative. The invention also provides a method for detecting the mercury ions, which comprises the step of quantitatively detecting the content of Hg in a solution of all water (pH = 7.4) by using the NBD derivative as a probe. The detection method can detect the mercury ions in an aqueous solution with high selectivity and sensitivity, and is simple, convenient and rapid to operate.Meanwhile, test paper for detecting the mercury ions is provided, so that the Hg detection is more convenient.

The invention discloses a benzooxaoxadiazole derivative as well as a preparation method and application thereof. The benzooxaoxadiazole derivative is prepared as follows: firstly, NBD-PZ is obtained by reaction between NBD-Cl and piperazine; then 4-nitro-7-(N-1'-acryloylpiperazine)-2,1,3-benzoxoxaoxadiazole is obtained by reaction between NBD-PZ and acryloyl chloride. The benzooxaoxadiazole derivative BGAS disclosed by the invention as a fluorescent probe has high selectivity for cysteine, can specifically detect the cysteine, particularly has a rapid detection effect on the cysteine in waterand simultaneously has relatively high sensitivity, thereby being applied to the detection of the cysteine in living cells.
