The invention provides disulfide-bond-containing long arm biotin and a preparation method thereof. The compound can be applied to the bio-detecting field, reduces influences of the steric effect, and improves sensitivity of detection. The structure formula of the disulfide-bond-containing long arm biotin is shown in the description; biotin is taken as a raw material, and is subjected to acylation and active ester synthesis through acylation ethanediamine and thiodipropionic acid to prepare the disulfide-bond-containing biotin.
