The invention relates to the technical field of molecular probes, and discloses an EGFR-targeting fluorescence quenching probe and a preparation method and application thereof. The fluorescence quenching probe structurally comprises a pharmacodynamic skeleton structure, a fluorescence quenching group and a fluorescence labeling group, wherein the fluorescence labeling group is selected from BODIPYFL dye; and the fluorescence quenching group is selected from 2, 4-dinitrophenol. The pharmacodynamic skeleton structure can be combined with EGFR protein in a targeting mode, and when the quenchinggroup in the molecular probe leaves, the fluorescence labeling group can conduct specific tracing and imaging; and by means of molecular imaging, the expression process of specific cells and moleculescan be observed in real time, a target spot is traced, and therefore disease development status evaluation and accurate detection of EGFR mutation typing of lung cancer patients are achieved at the molecular pathological level.

A general approach is presented for developing small molecule-based fluorogenic probes suitable for no-wash imaging of endogenous kinases in live cells. Probe 1, including a fluorophore-quencher system, was only turned on upon reacting with its target kinase Btk, and disclosed Btk's cellular location in live cells without any washing.
