- Magnesium tetrapyrazinoporphyrazines: Tuning of the pKa of red-fluorescent pH indicators

Magnesium(ii) tetrapyrazinoporphyrazines (TPyzPzs) are excellent red fluorophores (λF ~ 663 nm, ΦF ~ 0.53 in THF). In this work, a series of magnesium(ii) complexes of unsymmetrical TPyzPzs bearing one or two phenol substituents was prepared. Suitable substitutions on the phenolic moiety tuned its pKa in the range of 5.5 to 13. Deprotonation of the phenolic group at higher pH induced a strong donor (phenolate) in the macrocycle that led to pH-dependent quenching of the red fluorescence of these indicators. pH sensing was proved in water solutions after the incorporation of TPyzPzs into two delivery systems-microemulsions and liposomes. The latter also serves as a simple model of biomembranes. Finally, a wavelength-ratiometric probe was constructed by the incorporation of a TPyzPz indicator and lipophilic pH-nonsensitive BODIPY dye into liposomes. Synthetic precursors for TPyzPzs, substituted pyrazine-2,3-dicarbonitriles, also represent donor-acceptor systems and the pH-dependent changes in absorption spectra may be easily visible to the naked eye.

A new photoactive fluorinated diamine, 2-(4-trifluoromethylphenyl)-4,5- bis(4-(4-amino-2-trifluoromethylphenoxy)phenyl)imidazole (TFIA), was synthesized and used for preparation of novel fluorinated poly(amide-ether-imidazole)s (PAEIs) by using direct polycondensation of the diamine with various dicarboxylic acids. The resulting PAEIs were amorphous and had intrinsic viscosity [η] in the range of 0.68-0.80 dL/g. The weight average (M w) and number average (Mn) molecular weights of one of the polymer, measured by GPC, were 18,600 g/mol and 11,500 g/mol, respectively. These polymers were readily soluble in a variety of organic solvents and formed low-colored and flexible thin films via solution casting. Their values of cut-off wavelength (λ0) were in the range of 380-390 nm, and all PAEIs films exhibited high optical transparency. According to thermal analysis, these polymers exhibited glass transition temperatures (T g)s in the range of 160-250°C, initial decomposition temperature (Ti) from 295 to 400°C and temperature of 10% weight loss (T 10%) from 400 to 490°C in N2. These polymers exhibited strong UV-vis absorption at 310 nm in solution and in films and their fluorescence emission peaks appeared around 430-520 nm with the quantum yields in solution varied from 10% to 29%.

- Synthesis and evaluation of quinoxaline derivatives as potential influenza NS1A protein inhibitors

A library of quinoxaline derivatives were prepared to target non-structural protein 1 of influenza A (NS1A) as a means to develop anti-influenza drug leads. An in vitro fluorescence polarization assay demonstrated that these compounds disrupted the dsRNA-NS1A interaction to varying extents. Changes of substituent at positions 2, 3 and 6 on the quinoxaline ring led to variance in responses. The most active compounds (35 and 44) had IC50 values in the range of low micromolar concentration without exhibiting significant dsRNA intercalation. Compound 44 was able to inhibit influenza A/Udorn/72 virus growth.

Nineteen neutral nickel bisdiphenylethenedithiolene complexes [Ni(dpedt)2], including seventeen new compounds, were synthesized and characterized by a new short, efficient and multigram-scale synthetic method. Preliminary physical investigations show that these complexes are electron-withdrawing materials exhibiting good thermal stability and high molecular absorption coefficient in the near-infrared spectral region. Electrochemical and optical properties of [Ni(dpedt)2] depend strongly on the nickel bis(1,2-dithiolene) core and very slightly on the alkyl chain lengths or configurations. These highly soluble coordination compounds are potential candidates as acceptor solution-processable materials for active layers in organic electronic devices, such as field effect transistors or photovoltaic devices.

Three new low bandgap conjugated copolymers with 3,4-ethylenedioxythiophene (EDOT) as donor and 2,3-bis(4-octyloxyphenyl)-quinoxaline (P1), 2,3-bis(4-octyloxyphenyl)-thiadiazol-quinoxaline (P2, P3) as acceptors were synthesized by Stille cross-coupling reaction, and their optical and electrochemical properties were studied. These polymers exhibited optical bandgap of 1.77, 1.29 and 1.13 eV, for P1, P2 and P3, respectively. Photovoltaic cells with device configuration of ITO/PEDOT: PSS/Copolymer: PCBM (1:4 w/w)/LiF/Al were fabricated. The measurements revealed an open-circuit voltage (Voc) of 0.52 V, short-circuit current density (Jsc) of 3.24 mA/cm2 and power conversion efficiency (PCE) of 0.60% for P1, and showed a Voc of 0.33 V, Jsc of 2.11 mA/cm2, PCE of 0.39% for P2.

The complexing ability of compounds 5b and 5h, as the representatives of both groups of compounds, with alkali and alkali earth metal cations were measured by the solvent extraction method. The results showed that crown ether 5b comparatively had more affinity towards the Mg2+ cation, while thiacrown ether 5h had greater affinity towards the Ca2+ cation. A novel series of luminescent low molecular mass materials containing a 2,3-dicyanopyrazine central core were synthesized through an esterification reaction between diphenol 10 and different aromatic carboxylic acids 1-6, containing terminal long alkyl chains. They have a similar V-shaped geometry with lack of planarity between the two arms, confirmed by the X-ray structure of the central core. The optical and thermal properties of these compounds were evaluated. They show blue fluorescence in solution (λemmax 440-480 nm) with quantum fluorescence yields (ΦF) from 0.003 to 0.1 and Stokes shifts of around 90 nm. In solid state, optical band gaps (Eg) were from 3.14 to 3.32 eV. Thin films of 11, 13, and 14 exhibited blue fluorescence (λemmax 430-456 nm), and 12, 15, and 16 (more bulky) displayed green fluorescence (λemmax 488-512 nm). Most of the materials exhibited good thermal stability, exhibiting an amorphous glassy state after melting. Transparent amorphous films were easily obtained through spin coating and characterized by AFM analysis.

A self-polymerizable AB2 monomer, 2,3-bis(4-hydroxyphenyl)-6-fluoroquinoxaline, and an A2B monomer, 2,3-bis(4-fluorophenyl)-6-(4-hydrcixyphenoxy)quinoxaline) were prepared and polymerized to afford phenol-terminated and aryl fluoride terminated, hyperbranched polyphenylquinoxalines (HPPQs), respectively. MALDI-TOF analysis showed that intramolecular cyclization was a dominant process for the low molecular weight portion during the polymerizations. After isolation and complete dryness, the phenol-terminated HPPQ 1 was only soluble in strong organic acids, while the aryl fluoride terminated HPPQ 2 was soluble in most common organic solvents. HPPQ 1 was treated with allyl bromide to afford an allyl ether terminated HPPQ 3, which was also soluble in most organic solvents. Intrinsic viscosity measurements and SEC analysis indicated that HPPQ 2 had a much higher Mw and a much broader molecular weight distribution (PDI ~ 60) than HPPQ 1 and HPPQ 3 (PDI ~ 4). The results also suggested that HPPQ 1 formed aggregates in solution and that HPPQ 2 had a much more extended and open conformation. All the HPPQs, which were highly fluorescent, had UV absorption maxima near 375 nm in THF. However, the wavelength of their emission maxima, which ranged from 424 to 466 nm, depended on their end groups.

Two series of bis[1,2-bis(4-n-alkylphenyl)ethane-1,2-dithiolene]nickel, Cn-Ni (n= 1-12), and bis[1,2-bis(4-n-alkoxyphenyl)ethane-1,2-dithiolene]nickel, CnO-Ni(n = 1-12, 14, 16, 18), have been synthesized. Their mesomorphism, thermochromism, supramolecular structures and π-acceptor property have been investigated by using different scanning calorimetry, polarizing microscopy, temperature-dependent X-ray diffraction technique, electronic spectroscopy and cyclic voltammetry. From the X-ray diffraction and electronic spectral results, it was established that the CnO-Ni complexes for n ≤ 10 exhibit two differently colored discotic lamellar (DL) mesophases whereas none of the Cn-Ni complexes has a mesophase, and that the thermochromism (brown→green) is attributable to a slow transformation from the Ni-Ni bonded dimers to the Ni-S bonded dimers.
