The fluorescence emission intensity of the dansyl group is significantly diminished upon appending an ethyldimethylamino group to the N1 nitrogen substituent. Addition of acids and metal ions (i.e., Zn2+) to solutions of trimethylethylenediamine naphthalene sulfonamide (trinsyl) 2 produces a >25-fold increase in fluorescence intensity. Trinsyl probe 2 has been used as a diagnostic for the diffusion of protons and metal ions in a network polymer as well as an optical reporter for the glass transition temperature.

The inverse electron demand Diels–Alder pyridazine elimination reaction between tetrazines and allylic substituted trans-cyclooctenes (TCOs) is a key player in bioorthogonal bond cleavage reactions. Determining the rate of elimination of alkylamine substrates has so far proven difficult. Here, we report a fluorogenic tool consisting of a TCO-linked EDANS fluorophore and a DABCYL quencher for accurate determination of both the click and release rate constants for any tetrazine at physiologically relevant concentrations.

The present invention relates to peptides that are derived from HSV-2 glycoprotein G2 and represent HSV-2 type-specific epitopes. The present invention also provides for compositions comprising these peptides for type-specific serological diagnosis of HSV-2 infection. Methods of using these peptides for type-specific detection of HSV-2 antibodies and differentiation of HSV-2 viral infection from HSV-1 and other herpes family viral infections are further provided.

Provided are methods for evaluating male fertility based on the level of EP2D bound to sperm, kits for evaluating male fertility, and methods for enhancing or suppressing fertility in a male by regulating the level of EP2D and the binding between sperm and EP2D.
