- Synthesis and structure elucidation of hexetidin degradation products

Hexetidin, a hexahydropyrimidine derivative is present in many pharmaceutical formulations and exhibits an antimicrobial effect against a broad spectrum of microorganism. The synthesis of three degradation products of hexetidin which occur in aqueous formulations is subject of this publication. The structure and identity of these products is proven by means of NMR and IR studies. The synthesis route to yield triamin, hexedin and dehydrohexetidin provides evidence for a previously published disproportion theory.

- Stability and new formulations of hexetidine. Part 1: Analysis and stability

In aqueous formulations the antiseptic hexetidine hydrolizes in a first quick step for about 5% to an equilibrum. The further decomposition can be described by a second order kinetic. Comparing room-temperature studies with high-temperature stress tests, it can be demonstrated, that the Arrhenius equation is not valid. The stability optimum is at pH 5.5. The analytical investigations were realized by validated GC and HPLC methods.

- Analytical profile of purified hexetidine

Physico chemical, spectroscopic (UV, IR, NMR, mass), chromatographic (GLC, TLC) properties and synthesis of 1,3 bis(2 ethylhexyl) 5 amino 5 methyl hexahydropyrimidine (hexetidine) are reported and discussed. The difference between commercial and purified hexetidine is demonstrated.
