- Photophysical study of 6-amino-3-methyl-4-(4-nitrophenyl)-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile and estimation of ground-state and singlet excited-state dipole moments by solvatochromic approaches

An investigation of absorption and fluorescence behaviour of 6-amino-3-methyl-4-(4-nitrophenyl)-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile (NDPPC) at room temperature was carried out using a series of twelve organic solvents with different polarities. Ground and excited state dipole moments were determined experimentally by using Lippert–Mataga polarity function, Bakhshiev solvent polarity parameter, Kawski-Chamma-Viallet solvent polarity parameter, Reichardt's microscopic solvent polarity parameter, Kamlet-Abboud-Taft multiple linear regression and Catalan dipolar polarizibility approach. Due to considerable π- electron density redistribution, the value of excited state dipole moment was found to be greater than that of the ground state. The ground state dipole moment value was determined by quantum chemical method which was used to estimate excited state dipole moment using solvatochromic correlations. Both non-specific solute-solvent interactions and hydrogen bonding interactions were investigated by using Kamlet-Abboud-Taft and Catalan method. TD-DFT (B3LYP/6-311G (d,p)) was used for the determination of HOMO-LUMO energies.

Concentrated solar radiation (CSR)-assisted synthesis of pyranopyrazole derivatives under solvent and catalyst-free condition has been reported in the present protocol. One-pot multicomponent synthesis from aromatic/heteroaldehyde, ethyl acetoacetate, malononitrile, and hydrazine hydrate leads to the final desired compounds in a good yield. This operationally simple methodology has several advantages such as green and clean reaction profile, simple workup and purification procedures, extremely short reaction time, atom efficiency, and being economical. A novel task-specific ionic liquid, 2-carboxy-N,N-diethylethanaminium acetate denoted as [Et2NH(CH2)2CO2H][AcO], is prepared in an easy process and fully characterized by using suitable methods such as FT-IR, 1H-NMR, 13C-NMR, and TG, DTG and DTA analyses. After preparation, the promoting ability of this catalyst for one-pot multicomponent synthesis of 2-amino-4H-chromenes, dihydropyrano[3,2-c]chromenes and dihydropyrano[2,3-c]pyrazoles using in situ-developed arylidenemalononitriles through Knoevenagel reaction under solvent-free conditions is described. The sustainable procedure, short reaction times (6–36?min), effortless work-up, non-chromatographic purification, good-to-excellent product yields (79–96%), ease of recovery and reusability of the catalyst up to six reaction runs without a decrease of activity are the notable features of the present protocol.

For the first time, alpha-Casein was used as an efficient and eco-friendly catalyst for an effective and facile preparation of dihydropyranopyrazoles and spiropyranopyrazoles. The synthesis of bis (pyrazol-5-ols) derivatives was developed via one-pot, pseudo-five-component condensation, and the target dihydropyrano[2,3-c]pyrazoles and spiropyranopyrazoles were prepared by one-pot four-component reaction. This new method of employing alpha-Casein, which is a green, recyclable, non-toxic and commercially available catalyst, offers advantages such as mild condition, short reaction times, easy work-up, no need for column chromatography, and high yields of the products which make it more economic than other environmentally synthetic protocols.

Abstract: A clean and more economic protocol for the synthesis of pyrano[2,3-c]pyrazoles and pyrazolyl-4H-chromenes has been carried out using bael fruit ash (BFA) as a non-conventional natural catalyst in aqueous condition at ambient temperature. The catalyst was obtained from renewable resources by simple thermal treatment to dry rind of Aegle marmelos (Bael) fruit and formation of its active phase was confirmed by AAS, DSC-TGA, XRD, FT-IR, and SEM techniques. The BFA catalyst was found to be a green, highly active, easily biodegradable, and recyclable without loss of activity after the fifth run. To attain this goal , here, the present work synthesis, simulation, and antimicrobial studies of pyranopyrazole derivatives were discussed. Among the reported compounds, 4b has dominant antimicrobial activity that was due to its higher dipole moment. Also, this compound has the most hydrophilic nature and low Eg value. Besides, compound 4e has the next higher dipole moment, hydrophilic property and efficient biological activity. The rationale of these results explained that the comparison of electronic results with biological data is the better way to find the potent pharmaceutical drug compounds. A series of 6-amino-4-substituted-3-methyl-2,4-dihydropyrano[2,3-c]pyrazole-5-carbonitriles 5a–j were synthesized via one-pot, four-component condensation reactions of aryl aldehydes 1a–j, propanedinitrile (2), hydrazine hydrate (3) and ethyl acetoacetate (4) under solvent-free conditions. We report herein the use of the Br?nsted acid ionic liquid (BAIL) triethylammonium hydrogen sulphate [Et3NH][HSO4] as catalyst for this multi-component synthesis. Compared with the available reaction methodology, this new method has consistent advantages, including excellent yields, a short reaction time, mild reaction conditions and catalyst reusability. Selected synthesized derivatives were evaluated for in vitro anticancer activity against four human cancer cell lines viz. melanoma cancer cell line (SK-MEL-2), breast cancer cell line(MDA-MB-231), leukemia cancer cell line (K-562) and cervical cancer cell line (HeLa). Compounds 5b, 5d, 5g, 5h and 5j exhibited promising anticancer activity against all selected human cancer cell lines, except HeLa. Molecular docking studies also confirmed 5b and 5d as good lead molecules. An in silico ADMET study of the synthesized anticancer agents indicated good oral drug-like behavior and Non-toxic nature.

Chromenes, the oxygen-containing heterocyclic compounds, have a “special” place in biologically active natural products and in synthetic chemistry and in the fields of medicinal, agrochemical, cosmetic, and pigment industries. In this work, piperidine was used as a base catalyst for the convenient synthesis of 1,4-dihydropyrano[2,3-c]pyrazole, 5,10-dihydro-4H-benzo[g]chromene, and pyrazolo[1,2-a] [1,2,4]triazole derivatives at ambient temperature. This methodology has several advantages including the use of easily accessible and inexpensive catalysts, short reaction times, high yields, convenient work-up, and not needing column chromatography.

Abstract: This article aimed to present two facile and environmental friendly routes for the rapid assembly of?biologically active compounds including pyrazol core using aspirin as a novel and green catalyst. The synthesis of bis(pyrazol-5-ol) derivatives was developed via one-pot, pseudo-five-component condensation, and the target dihydropyrano[2,3-c]pyrazoles and spiropyranopyrazoles were prepared by one-pot, four-component reaction. These reactions can be performed in tandem from readily available starting materials. The main merits of the present methods are operational simplicity, no need for column chromatography, inexpensive materials, avoidance of harmless and corrosive acid catalysts, short reaction times, good yields of the products, and utilization of aspirin as a non-toxic, cheap, commercially available, and efficient catalyst.
