  • General Description (17β)-2,4-Dinitroestra-1(10),2,4-triene-3,17-diol (also known as 2,4-dinitroestradiol) is a nitrated derivative of estradiol formed under oxidative/nitrosative stress conditions via peroxidase-mediated nitration. It is produced alongside 2-nitroestradiol and 4-nitroestradiol, with its formation favored at higher estradiol concentrations (10–300 μM) but negligible at physiological levels (e.g., 1 μM). (17beta)-2,4-dinitroestra-1(10),2,4-triene-3,17-diol arises from further nitration of 4-nitroestradiol, which exhibits greater susceptibility to nitration than its 2-nitro counterpart. While 2,4-dinitroestradiol itself was not directly tested for biological activity, related nitroestradiols (e.g., 2-nitroestradiol) demonstrated adverse effects on bovine embryo development, suggesting potential hormonal dysfunction. However, the physiological relevance of such nitration products remains uncertain due to their low yields under biologically plausible conditions.
