The research presents an improved synthesis method for [18F]fluoromethyl tosylate, a reagent crucial for radiofluoromethylation processes. The study reexamined the preparation of this compound through the reaction of bis(tosyloxy)methane with 18F-, which was reported to yield [18F]fluoromethyl tosylate along with [18F]tosyl fluoride. The experiments involved various reaction conditions, including the addition of water, to optimize the yield of [18F]fluoromethyl tosylate, which was significantly increased to 71±6% with the defined reaction conditions. The analyses used in the study included thin layer chromatography (TLC) and high-performance liquid chromatography (HPLC) to monitor the product formation and purity. The synthesized [18F]fluoromethyl tosylate was then used to produce [18F]fluticasone propionate and other radiolabeled compounds, demonstrating its utility as a labeling reagent.
