A Facile, One-Pot Synthesis of 2-Arylthiazolo<5,4-b>pyridines

The research describes a novel and effective one-pot synthesis method for 2-arylthiazolo[5,4-b]pyridines, a class of fused heterocycles known for their analgesic, antipyretic, and anti-inflammatory properties. The key chemicals involved in this synthesis include 3-amino-2-chloropyridines (1a,b), which are commercially available, and O-ethyl arene- or heteroarenethiocarboxylates (2a-d), which can be prepared either by reacting the corresponding ethyl carboxylates with Lawesson reagent in toluene or by converting aromatic or heteroaromatic nitriles into imidic esters via Pinner reaction and subsequent treatment with hydrogen sulfide. The synthesis process involves condensation of these thioesters with the Grignard reagent of 2-chloro-3-aminopyridines in tetrahydrofuran, yielding 2-arylthiazolo[5,4-b]pyridines (3a-h) with good yields. The necessity of a chlorine substituent on the pyridine ring is highlighted through comparison with reactions involving 3-aminopyridine. The study provides detailed procedures, yields, melting points, and spectral data for the synthesized compounds.
