The researchers optimized the reaction conditions, finding that treating aldehydes with DBI in dichloromethane at room temperature efficiently generates acyl bromide intermediates. These intermediates then react with various amines, including primary, secondary, and cyclic amines, to produce the desired amides in high yields (81–94%) without significant side products. The study highlights the practicality of this method by demonstrating its scalability through a gram-scale reaction, yielding the desired product in 90% yield. This metal-free, mild condition oxidative amidation offers a simple, efficient, and environmentally friendly alternative for the direct synthesis of amides from aldehydes, with potential applications in pharmaceuticals and other industries.
