The invention relates to a terminal-hydroxyl organic phosphorus-nitrogen-sulphur ester content and a preparation method thereof and flame-retardant polyester fiber POY(polyester pre-oriented yarn) made from the terminal-hydroxyl organic phosphorus-nitrogen-sulphur ester content. Firstly, DAPER with terminal being hydroxyl is obtained through reaction of lateral-group phosphorous dicarboxylic acid with pentaerythritol, then DADA with one end being sulfydryl while is obtained through reaction of terminal-hydroxyl nitrogen contained monocarboxylic acid with dihydric alcohol, and finally, a final product is obtained through reaction of the DAPER with the DADA. The final product is of the solid ester content with high molecular weight, the terminal of the molecular is the hydroxyl, 15.1-16.04wt% of phosphorus, nitrogen and sulphur are contained, and the ester content has the advantages of high thermal stability, wide application and the like; the final product is taken as a flame retardant to prepare the flame-retardant polyester fiber PLY, limit oxygen index of the flame-retardant polyester fiber POY is greater than or equal to 38%, filament number is from 0.3dtex to 1.0dtex, and breaking strength is greater than or equal to 2.1cN/dtex.
