EIPA, also known as 5-(N-ethyl-N-isopropyl)-amiloride, is a potent inhibitor of sodium-hydrogen exchangers (NHE), which are involved in maintaining sodium and pH balance in various tissues. It selectively inhibits NHE1, NHE2, NHE3, and NHE5 isoforms with Ki values of 0.02, 0.5, 2.4, and 0.42 μM, respectively, and less effectively inhibits NHE4 (IC50 ≥10 μM). EIPA is commonly used at a concentration of 5-10 μM to inhibit cellular NHE activity and is a yellow solid.
Used in Biological Research: EIPA is used as a selective Na+/H+ antiporter inhibitor for studying the role of NHE in various biological processes. It selectively enhances the excretion of sodium ions without causing an increase in the excretion of potassium ions. Used in Cell Culture: EIPA is used as a supplement in endothelial cell growth medium to incubate human lung microvascular endothelial cells in Pluronic F-127 stock solution. Used in Drosophila Larval Live-Imaging: EIPA is used in choline chloride solution as an experimental perfusate to study the effect of Na+/H+ inhibition on jejunal loop perfusion in vivo. Used in In Vivo Studies: EIPA is used as an experimental perfusate in choline chloride solution to study the effect of Na+/H+ inhibition on jejunal loop perfusion in vivo.
