Synthesis and Pharmacological Evaluation of N1--N4-arylthiosemicarbazides and 2-Arylamino-5--1,3,4-thiadiazoles/oxadiazoles as Oral Hypoglycemic Agents

A series of 2-arylamino-5--1,3,4-thiadiazoles/oxadiazoles (III/IV) have been synthesised from N1--N4-arylthiosemicarbazides (II) by treatment with conc. sulphuric acid or mercuric oxide.The thiosemicarbazides (II), in turn, have been prepared by refluxing 2--3-arylquinazolin-4(3H)-one (I) with various aryl isothiocyanates in abs. ethanol.A few of these compounds when tested on rats at an oral dose of 250 mg/kg, reduced the blood sugar to about 25 percent.
