The present invention provides a class of 2-(5,6-methylxanthone-4-yl)-acetic acid derivatives with a novel structure, wherein a structure of a general formula is represented by a picture. The present invention further provides a method for preparing the derivatives, wherein multiple structure modification reforming are performed on a carboxyl portion of 2-(5,6-methylxanthone-4-yl)-acetic acid to prepare the derivatives. Compared with the existing 2-(5,6-methylxanthone-4-yl)-acetic acid or derivatives thereof, a part of the derivatives prepared by using the method of the present invention provide inhibition effects for a variety of tumor cells, particularly provide high tumor cell inhibition rate for lung cancer and leukemia, and show significant anti-tumor activities. According to the present invention, the design is rational, sources of the raw materials for preparation are wide, the raw materials are easy to obtain, the preparation method is simple and easy to operate, reaction conditions are mild, the product yield is high, and the method is applicable for industrial large scale production.
