ONE-POT SYNTHESIS OF SOME FUSED PYRAN-2-ONES

A one-pot synthesis of different fused pyran-2-ones starting from some less commonly used activated methylene compounds (cyclic lactones, a thiolactone and a lactam), one-carbon sources and N-acylglycines in acetic anhydride is described.

Methyl 2-benzoylamino-3-dimethylaminopropenoate (1) reacts in acetic acid with monocyclic or bicyclic heterocyclic compounds with two hydroxy or potential hydroxy groups in 1,3-position to give fused pyranones.Accordingly, derivatives of 2H-pyranopyridine (3), 2H-pyranoquinoline (9, 10, and 11), 2H-pyranopyridazine (13), 8-H-pyranotetrazolopyridazine (16), pyranopyran (18), and 2H-pyranopyridine (22) were obtained.

Treatment of 4-ethoxymethylene-2-phenyl-5(4H)-oxazolone with activated methylene compounds under acidic or basic conditions leads to 2H-pyran-2-ones and fused pyran-2-ones.On the other hand, methyl (3-benzoylamino-5-methoxycarbonyl-2-oxo-2H-pyran-6-yl)acetate (4) has also been prepared by a one-pot synthesis from dimethyl 1,3-acetone dicarboxylate, diethoxymethyl acetate, hippuric acid and acetic anhydride.
