The invention discloses a 3-quaternary carbon chiral-C-acylbenzofuranone derivative and a preparation method thereof. O-acylbenzofuranone derivative as a raw material undergoes an asymmetric Black rearrangement reaction in the presence of a chiral imidazole catalyst to produce the 3-quaternary carbon chiral-C-acylbenzofuranone derivative. The preparation method utilizes a cheap and easily available chiral nucleophilic catalyst to replace an expensive catalyst used in the prior art, has mild catalytic reaction conditions and simple processes, and has a good yield and enantioselectivity. Through further derivation, the obtained 3-quaternary carbon chiral-C-acylbenzofuranone derivative can be used for synthesis of a physiologically active molecule.
