Arene diruthenium(II)-mediated synthesis of imines from alcohols and amines under aerobic condition

The utility and selectivity of the newly synthesized dinuclear arene Ru(II) complex were demonstrated towards the synthesis of imines from coupling of alcohols and amines in the aerobic condition. Analytical and various spectral methods have been used to establish the unprecedented formation of the new thiolato-bridged dinuclear ruthenium complex. The molecular structure of the titled complex was evidenced with aid of X-ray crystallographic technique. A wide range of imines were obtained in good-to-excellent yields up to 98% and water as the by-product through an acceptorless dehydrogenative coupling of alcohols with amines. The catalytic reaction operated a concise atom economical without any oxidant with 1 mol% of the catalyst load. Further, the role of base, solvent and catalyst loading of the coupling reaction has been investigated. A plausible mechanism has been described and was found to proceed via the formation of an aldehyde intermediate. Short synthesis of antibacterial drug N-(salicylidene)-2-hydroxyaniline illustrated the utility of the present protocol.

The occurrence of resistances in Gram negative bacteria is steadily increasing to reach extremely worrying levels and one of the main causes of resistance is the massive spread of very efficient β-lactamases which render most β-lactam antibiotics useless. Herein, we report the development of a series of imino-analogues of β-lactams (namely azetidinimines) as efficient non-covalent inhibitors of β-lactamases. Despite the structural and mechanistic differences between serine-β-lactamases KPC-2 and OXA-48 and metallo-β-lactamase NDM-1, all three enzymes can be inhibited at a submicromolar level by compound 7dfm, which can also repotentiate imipenem against a resistant strain of Escherichia coli expressing NDM-1. We show that 7dfm can efficiently inhibit not only the three main clinically-relevant carbapenemases of Ambler classes A (KPC-2), B (NDM-1) and D (OXA-48) with Ki's below 0.3 μM, but also the cephalosporinase CMY-2 (class C, 86% inhibition at 10 μM). Our results pave the way for the development of a new structurally original family of non-covalent broad-spectrum inhibitors of β-lactamases.

We report the synthesis of β-lactams from α-ketoacylsilanes and imines, which proceeds via a formal [2+2] photochemical cycloaddition with in situ generation of siloxyketene. This mild and operationally simple reaction proceeds in an atom-economic fashion with broad substrate scope, including aldimines, ketimines, hydrazones, and fused nitrogen heterocycles, affording a variety of important β-lactams with satisfactory diastereoselectivities in most cases. This reaction also features good functional-group tolerance, facile scalability and product diversification. Experimental and computational studies suggest that α-ketoacylsilanes can serve as photochemical precursors by engaging in a 1,3 silicon shift to the distal carbonyl group.

Three copper(I) complexes, [CuCl(L)(PPh3)2] [L = FL (1), BL (2) or TL (3)] were prepared from [(PPh3)2Cu(μ-Cl)2Cu(PPh3)] and N-carbamothioylfuran-2-carboxamide (FL), N-carbamothioylbenzamide (BL) or N-carbamothioylthiophene-2-carboxamide (TL) ligands in benzene and four-coordinated tetrahedral copper complexes were well characterized by various spectroscopic techniques (UV/Vis, FT-IR, 1H NMR, 13C NMR and 31P NMR). The molecular structure of the ligands (FL and BL) and complexes was established from single-crystal X-ray diffraction studies. Copper complexes have been shown to catalyse the one-pot synthesis of imines and quinoxalines. Heterogenized catalyst (4) was prepared by reacting more active complex 3 with polystyrene supported triphenylphosphane, and characterized by elemental analyses, and DRS-UV, FT-IR, ICP-OES, and solid-state NMR techniques. Catalytic activity of the complexes (3 and 4) was tested in the formation of imines from alcohols and amines, and quinoxalines from hydroxy ketones and diamines. Heterogeneity and reusability of catalyst 4 were evaluated, and the catalyst can be reused for four runs without any loss in activity.

Oxidation of aromatic anils in aniline moiety by meta-chloroperbenzoic acid in aqueous acitic medium

The kinetics of oxidation of aromatic anils in aniline moiety by meta-chloroperbenzoic acid (m-CPBA) has been studied in aqueous acitic medium. The order of reaction was found to be second order with respect to aromatic anil and first order with respect to m-chloroperbenzoic acid. The increase of [H+] in this oxidation retards the rate of reaction. The effects of substituents on the oxidation rate were studied with 3 meta- and 5 para-substituted anils at five different temperatures. Thermodynamic parameters for the oxidation have been determined and discussed. meta-Chloroperbenzoic acid oxidation with substituted anils fulfills the isokinetic relationship and Exner relationship but not to any of the linear free energy relationships. The deviation of Hammett plot was noted and a concave downward curve was obtained for the anils with substituents in aniline moiety.

Oxidation of aromatic anils by sodium perborate in aqueous acetic Acid Medium

The kinetics of oxidation of 9 meta- and 15 para- substituted aromatic anils by sodium perborate were investigated in aqueous acetic acid medium. The reaction was second order with respect to aromatic anil and first order with respect to the sodium perborate. The increase of [H+] in this oxidation retards the rate of the reaction. The observed rate constant for the substituents were plotted against the Hammett constant, δ and a non-linear concave downward curve was obtained for the anils with substituents in the aniline moiety. The observed break in the log kobs versus δwas attributed to the transition state whereas the non-linear concave upward curve was observed for the substituents in the benzaldehyde moiety and a non-linear concave upward curve was observed for the substituents in the combination of aniline and benzaldehyde moiety. The electron withdrawing substituents fall on one side of the curve, having a negative ρvalue and the electron releasing substituents fall on the other side, with a positive ρvalue and a suitable mechanism was proposed.

The development of a novel intermolecular oxidative C-H alkoxylation of aniline derivatives is described under metalfree conditions with high reaction rates at ambient temperature. In the presence of an I(III) oxidant, a range of aldehydes, anilines, and alcohol substrates undergo three-component coupling to produce synthetically useful alkoxyl-substituted N-arylimines. The preliminary mechanism investigations revealed that the transformation proceeds via imines as intermediates.
