The measurement of serum/plasma levels of 1α,25-dihydroxyvitamin D3 1 a is important for the diagnosis influencing vitamin D metabolism.To obtain antibodies to the metabolite 1a which are highly specific and useful for development of immunoassays, two novel haptenic derivatives, 11α-(3-carboxypropanoyloxy)-1α,25-dihydroxyvitamin D3 2a and 11α-(4-carboxybutanoyloxy)-1α,25-dihydroxyvitamin D3 2b were synthesized each in 19 steps from a suitably protected derivative (3) of 11α,25-dihydroxycholesterol.

The production of highly specific antibodies to 1α,25-dihydroxyvitamin D3 la utilizing a novel haptenic derivative 2, which was synthesized from 11α,25-dihydroxycholesterol via 21 steps, has been reported. The properties of the obtained antibodies are also described.
