Usage

Description

Fomivirsen, also known as Fomivirsen Sodium or Vitravene, is a phosphorothioate oligonucleotide (PS) consisting of a sequence of 21 nucleotides. It is synthesized using solid-phase phosphoramidite chemistry and functions as an antisense inhibitor, targeting an immediate early mRNA of cytomegalovirus (CMV) to inhibit the expression of two regulatory proteins. Fomivirsen was the most advanced first-generation PS drug candidate in human clinical trials and was the first agent based on antisense technology to reach the market.

Uses

Used in Ophthalmology:
Fomivirsen is used as a local treatment for CMV-induced retinitis in AIDS patients. It is administered through intravitreal injections to inhibit human CMV replication, which can cause severe vision loss or blindness in these patients. Fomivirsen has demonstrated the ability to inhibit AD169 human CMV replication by more than 90% at a concentration of 5μM, showing greater inhibitory properties than ganciclovir in several experimental models.
Used in Antiviral Therapy:
Fomivirsen is used as an antiviral agent for the treatment of cytomegalovirus infections, particularly in immunocompromised patients such as those with AIDS. Its antisense mechanism of action allows it to specifically target and inhibit the replication of CMV, making it a valuable tool in the fight against viral infections in vulnerable populations.
