The invention relates to a synthesis method of monomethine cyanine dye. The method comprises the following steps: (1) adding N-methyl quinoline-2-thioketone, methyl p-toluenesulfonate, active methyl-containing heterocyclic quaternary ammonium salt and absolute ethyl alcohol into a round-bottom flask; dropwise adding triethylamine; and performing reflux; and (2) performing reduced-pressure distillation on the solution after the reflux in the step (1) to separate out partial solvent, and cooling; and when solid is separated out, performing suction filtration, drying and recrystallizing to finally obtain a finished product of monomethine cyanine dye. The method provided by the invention has the advantages of easiness in operation, high synthesis yield, a few steps and short time, can effectively avoid the purifying treatment of the intermediate product, and realizes high synthesis efficiency and relatively low synthesis cost.
