The invention provides a synthetic method of 2-amidogen-4-chlorine-5-fluorine pyrimidine. The synthetic method of the 2-amidogen-4-chlorine-5-fluorine pyrimidine comprises the steps that 2,4-dichloro-5-fluorine pyrimidine is taken and dissolved in tetrahydrofuran, reducing to a low temperature is carried out, ammonium hydroxide is added, and low-temperature reaction is kept for 2-3h after the ammonium hydroxide is added; rising to a room temperature naturally is carried out and reaction is carried out for 1-2h, and a mixture of 2-chlorine-4-amidogen-5-fluorine pyrimidine and the 2-amidogen-4-chlorine-5-fluorine pyrimidine is obtained; and the obtained mixture is subjected to column chromatography isolation to obtain the 2-amidogen-4-chlorine-5-fluorine pyrimidine. According to the synthetic method of the 2-amidogen-4-chlorine-5-fluorine pyrimidine, market 2,4-chlorine-5-fluorine pyrimidine is used as a raw material, and the 2-amidogen-4-chlorine-5-fluorine pyrimidine is generated through the substituted one-step reaction; the raw materials are greatly supplied by commercialization, are price and easy to obtain, ingredients are recycled and reused, the cost is obviously reduced, meanwhile, pollution of toxic substances to the environment in the production process is avoided, and the safety in the operation process is improved; and the yield is greatly improved, time is effectively shortened, the operation is simple, and the postprocessing process is simplified.

Also described herein are methods of using such Olig2 inhibitors, alone and in combination with other compounds, for treating cancer and other diseases. In particular the Olig2 inhibitors may be used to treat glioblastoma.
