The invention provides a synthetic method of 2-amidogen-4-chlorine-5-fluorine pyrimidine. The synthetic method of the 2-amidogen-4-chlorine-5-fluorine pyrimidine comprises the steps that 2,4-dichloro-5-fluorine pyrimidine is taken and dissolved in tetrahydrofuran, reducing to a low temperature is carried out, ammonium hydroxide is added, and low-temperature reaction is kept for 2-3h after the ammonium hydroxide is added; rising to a room temperature naturally is carried out and reaction is carried out for 1-2h, and a mixture of 2-chlorine-4-amidogen-5-fluorine pyrimidine and the 2-amidogen-4-chlorine-5-fluorine pyrimidine is obtained; and the obtained mixture is subjected to column chromatography isolation to obtain the 2-amidogen-4-chlorine-5-fluorine pyrimidine.
