The synthesis of haptens and properties of catalytic antibodies designed to catalyse carbanionic cyclisation reactions

The design of haptens to generate antibodies with the potential to catalyse carbanionic cyclisation reactions is introduced in the context of Dieckmann cyclisations and related reactions. Syntheses of five-membered ring haptens containing a sulphone, to represent the transition state(s) associated with the tetrahedral intermediate, and an amino group, to promote the formation of a general base, are described. Results with partially purified antibodies are reported showing that the target reaction can be catalysed by antibodies.
