The present invention is directed to oxazolobenzimidazole derivatives which are potentiators of metabotropic glutamate receptors, particularly the mGluR2 receptor, and which are useful in the treatment or prevention of neurological and psychiatric disorders associated with glutamate dysfunction and diseases in which metabotropic glutamate receptors are involved. The invention is also directed to pharmaceutical compositions comprising these compounds and the use of these compounds and compositions in the prevention or treatment of such diseases in which metabotropic glutamate receptors are involved.

The synthesis and structure-activity relationships (SAR) of Chk1 inhibitors based on a 5,10-dihydro-dibenzo[b,e][1,4]diazepin-11-one core are described. Specifically, an exploration of the 7 and 8 positions on this previously disclosed core afforded compounds with improved enzymatic and cellular potency.

Dibenzofurans, a dibenzothiophene, and carbazoles, each substituted with a 2-benzoxazolyl group as well as an ortho-hydroxyl group, were synthesized to produce fluors with fluorescence due to excited-state intramolecular proton-transfer. The orientations for Friedel-Crafts acylation of 3-methoxydibenzothiophene and of the analogous carbazole were determined. The fluors displayed absorption peaks in the 330-385 nm region with molar extinction coefficients up to 57,000. Fluorescence quantum efficiencies of 0.17-0.44 were obtained at wavelengths that had peak values from 540-600 nm. The fluors are of potential use as wavelength shifters in scintillating polystyrene fibers.
