Synthesis of isotopically labelled 2-isopropylthioxanthone from 2,2′-dithiosalicylic acid and deuterium cumene

Two efficient synthetic routes of stable deuterium labelled 2-isopropylthioxanthone were presented with 98.1% and 98.8% isotopic abundance in acceptable yields and excellent chemical purities. Their structures and the isotope-abundance were confirmed according to proton nuclear magnetic resonance and liquid chromatography–mass spectrometry.

The invention relates to stable isotopic labeled 2-isopropylthioxanthone and a synthetic method thereof. The stable isotopic labeled 2-isopropylthioxanthone is 2-isopropylthioxanthone labeled by D or 13C. The synthetic method comprises the following steps of using benzene and acetone which are labeled by stable isotopic D or 13C as raw materials; reacting the benzene and the acetone with magnesium to obtain Grignard reagent after benzene bromination; performing reduction and hydroxide radical bromination on the acetone to obtain 2-bromic propane; performing Grignard reaction to obtain isopropyl benzene; reacting the isopropyl benzene with dithio-salicylic acid; and synthesizing to obtain the 2-isopropylthioxanthone labeled by the stable isotopic D or 13C. Compared with the prior art, the stable isotopic labeled 2-isopropylthioxanthone has the advantages that a process route is simple, the 2-isopropylthioxanthone is easy to synthesize, products are easy to separate and purify, the chemical purity of the products is above 99.0%, isotope abundance is above 98.0% atom, requirements of trace detection in the field of food safety can be met fully, and economical efficiency and using value are excellent.

The scope and mechanism of a practical protocol for the iron-catalyzed hydrogenation of alkenes and alkynes at 1 bar H2 pressure were studied. Intrinsic Deuterium Isotope Effects of Deuteriated tert-Butyl Groups on the 13C NMR Spectra of Aromatic Compounds

The 3Δ-deuterium isotope effects of partially and fully deuteriated tert-butyl groups on the 13C NMR spectra of tert-butylbenzene and derivatives are discussed in detail.It is shown that they correlate with the chemical shift of C-1 of the aromatic ring.It has been demonstrated that when deuterium is replaced with some other substituents, the SCS values of these substituents show a parallel behaviour to the deuterium isotope effects.It is concluded that, for the compounds studied, deuterium isotope effects and substituent chemical shifts can be described on a common basis.

The preparation of 4-fluoro-2-nitrobenzonitrile (V), an intermediate in the synthesis of the title compound I, from 4-fluoro-2-nitroaniline via 5-fluoro-2-iodonitrobenzene (VII) was elaborated.Syntheses of 1,1,1,3,3,3-hexadeutero-2-propyl (XX) and 1,3,4-trideutero (XXVIII) analogues of compound I from hexadeuteroacetone, and pentadeuterobromobenzene, respectively, were carried out.Compound I was esterified with acetic anhydride, decanoic acid and 3,4,5-trimethoxybenzoyl chloride to give the esters II-IV.Acylation of compound XXX with acetyl chloride, 4-fluorophenoxyacetyl chloride and (4-fluorophenylthio)acetyl chloride and the following reduction of the amides with lithium aluminium hydride gave compounds XXXII, XXXIX, and XL.Substitution reactions of 11-chloro-7-fluoro-2-isopropyl-10,11-dihydrodibenzothiepin with the corresponding N-monosubstituted piperazines resulted in compounds XXXIII-XXXV, XXXVII, XXXVIII, XLI and XLII.Alkylation of XXX with 2-(2-chloroethyl)-1,3-dioxolane afforded compound XXXVI.Pharmacological testing of the new compounds, derivatives and analogues of the neuroleptic agent isofloxythepin (I), for discoordinating and cataleptic activities, showed especially for compounds II, XXXIV and XXXVI very intensive and long-lasting effects.The decanoate III has properties of a depot neuroleptic agent.
