Antinociceptive properties of some 4-benzylpiperidine analogues of phencyclidine and thienylcyclidine

The 4-benzyl-1-(1-phenylcyclohexyl)piperidine, 4-benzyl-1-[1-(2- thienyl)cyclohexyl]-piperidine and the corresponding cis-3-methylcyclohexyl and cis-4-methylcyclohexyl analogues have been synthesized through the Bruylants reaction. The title compounds were characterized and their antinociceptive activity tested in mice by the hot-plate and writhing tests and compared with that of pethidine. The effect of naloxone on antinociceptive activity is also described. Although the Bruylants reaction is known to give exclusively cis products, the relative stereochemistry of the new diastereoisomers was further confirmed by using 13C-NMR and chemical shift correlation with spectra of closely related compounds of established stereochemistry.
