The invention discloses a method for preparing L-idose from monoacetone glucose as a raw material. The method comprises the following steps of S1, preparing 1, 2-O-isopropylidene-3-O-acetyl-alpha-D-glucofuranose; S2, preparing 1, 2-O-isopropylidene-3-O-acetyl-5, 6-di-O-p-toluenesulfonyl-alpha-D-glucofuranose; S3, preparing 1, 2-O-isopropylidene-3-O-acetyl-5, 6-di-O-benzoyl-beta-L-idose; S4, preparing 1, 2-O-isopropylidene-L-idose, adding 100-200 mL of a newly prepared sodium methoxide-methanol solution of 0.1 M into a product obtained in the step S3, performing stirring at the normal temperature for 30-90 min, adding cation exchange resin to remove Na ions, performing filtering, and concentrating a filtrate under reduced pressure; and S5, preparing the L-idose. An isomerization method adopted by the process route is mature and stable in process route, mild in reaction, small in environmental pollution caused by raw materials and reagents and relatively low in cost, an intermediate anda target product can be purified through a recrystallization method, the yield is relatively high and stable, and the method is suitable for industrial production.
