A series of pyran 1–6 and polyhydroquinoline derivatives 7–9 are synthesized targeting anticancer agents for clinical trials. Their structures are characterized by IR, 1H NMR, and HRMS spectra, and single crystal X-ray crystallography. Their anticancer activity against four human osteosarcoma cells (Saos-2, MG-63, 143B, and U2-OS) is evaluated in vitro using the MTT assay. The compounds 7–9 demonstrate high anticancer activity against the four cancer cells.

A clean, efficient and mild method is presented for one-pot three-component synthesis of 2-amino-7,7-dimethyl-4-(4-(methylthio)phenyl)-5-oxo-5,6,7,8-tetrahydro-4H-chromene-3-carbonitrile using nickel ferrite magnetic nanoparticles as a recyclable catalyst in green solvent at 40?°C. The structure of the target product was confirmed by FT-IR, 1HNMR, 13CNMR and single crystal X-ray structure analysis. In the present work, the quantum theoretical calculations of the molecular structure of this new compound have been predicted using Density Functional Theory in the solvent DMSO. The geometries of the title compound were optimized by B3LYP/6-31+G* and B3LYP/6-311++G** levels of theory. The electronic spectrum of the title compound in the solvent DMSO was carried out by TDB3LYP/6-311++G** method. Frontier molecular orbitals, molecular electrostatic potential, electronic properties, natural charges and natural bond orbital analysis of the mentioned compound were investigated and discussed by theoretical calculations. Taurine (2-aminoethanesulfonic acid), a semi-essential amino acid that exists in the human body and numerous other living creatures, is used as a green bio-organic catalyst for the promotion of the Knoevenagel reaction between aldehydes and malononitrile. In the same way, tetraketones can also be produced through a Knoevenagel reaction, followed by Michael addition. 2-Amino-3-cyano-4H-pyran derivatives are simply prepared via a three-component reaction in the presence of taurine as the catalyst. All these reactions are performed in water, a green solvent. The advantages of using of taurine as the catalyst are it is environmentally friendly, low cost, commercially available, easy to separate from the reaction mixture, and has high reusability. Use of this catalyst results in acceptable reaction times, high yields and high purities of the obtained products without utilizing any organic solvents.
