Phenolic Berbine Alkaloids: Total Synthesis of (+/-)-Discretine

A convenient total synthesis of (+/-)-discretine is described.Condensation of 6,7-dimethoxyisochroman-3-one (1) with 3-benzyloxy-4-methoxy-β-phenylethylamine (2) gives the phenylacetamide (3), which on cyclisation with PCl5, followed by NaBH4 reduction gives (+/-)O-benzyldiscretine (4).Debenzylation of 4 with ethanolic-HCl provides (+/-)-discretine (5) in high yield.
