Uses

Used in Medical Applications:
Protein C is used as a therapeutic agent for the treatment of severe sepsis and associated acute organ dysfunction. It acts as an antithrombotic by inhibiting factors Va and VIIIa, promotes fibrinolysis by inactivating plasminogen-activator inhibitor-1, and exerts anti-inflammatory effects by inhibiting the production of inflammatory cytokines.
Used in Preeclampsia Treatment:
Protein C is used as a treatment for severe early-onset preeclampsia in pregnant women, as it helps in prolonging pregnancy and improving perinatal outcomes.
Used in Laboratory Research:
Protein C from human plasma is used for the pre-treatment of endothelial cells prior to antibody-inhibition assays. It is also utilized in activated protein C (APC) assays to determine its inhibitory effect on copper.
Used in Drug Development:
Drotrecogin alfa, a recombinant form of activated protein C, was introduced as an intravenous treatment for reducing mortality in adult patients with severe sepsis. It is the first recombinant activated protein C expressed in human kidney 293 cells and is marketed under the brand name Xigris. However, it should be noted that the use of drotrecogin alfa has been associated with an increased risk of serious bleeding.

APC mutations cause accumulation of β-catenin in the nucleus leading to activation of LEF-1 and/or TCF, and the induction of target genes such as the oncogene c-myc. It is involved in regulation of mitotic chromosome separation and stability.
