Rhodium-catalyzed [2 + 2 + 2] cycloadditions, sulfonyl phthalide annulations and nitroalkene reactions have been employed for the synthesis of 56 quinone-based compounds. These were evaluated against Trypanosoma cruzi, the parasite that causes Chagas disease. The reactions described here are part of a program that aims to utilize modern, versatile and efficient synthetic methods for the one or two step preparation of trypanocidal compounds. We have identified 9 compounds with potent activity against the parasite; 3 of these were 30-fold more potent than benznidazole (Bz), a drug used for the treatment of Chagas disease. This article provides a comprehensive outline of reactions involving over 120 compounds aimed at the discovery of new quinone-based frameworks with activity against T. cruzi.

Hauser–Kraus annulation of sulfonylphthalides with simple conjugated nitroalkenes follows the expected pathway to furnish naphthoquinones in moderate yields. However, a strategic variation of this reaction by employing nitroalkenes bearing an additional nucleophilic site results in [4+4] annulation leading to complex fused and spiro heterocycles in high yields with high selectivity through a cascade process involving Michael addition, Dieckmann cyclization, and a series of eliminations and rearrangements.

1,3-Dithienylisothianaphthene (10a) is obtained through ring closure of 1,2-dithienoylbenzene (7a). The synthesis of 7a has been accomplished based on a Grignard reaction by adding 2-thiophene-magnesium bromide to 1,2-di(S-(2-pyridinyl)) benzenedithioate (6b) to obtain 7a in a yield of 95%. The use of 6b avoids the formation of the corresponding 3,3-dithienyl-3H-isobenzofuran-1(3H)-one (dithienylphthalide, 8). The same procedure is applied to obtain 1,3-dithienyl-4,5,6,7-tetradeuterioisothianaphthene (10b) and 1,3-dithienyl-4,5,6,7-tetrafluoroisothianaphthene (10c). The synthesis of the 2,3-dithienoylpyridine (12a), 3,4-dithienoylpyridine (12b), and 2,3-dithienoylpyrazine (12c) however fails. The presence of nitrogen in the central ring system influences the result of the Grignard reaction. Possibly the free electron pair of the nitrogen interferes with the formation of a stable six-membered ring intermediate which is essential for the diketone formation.

(Acyloxy)alkyl α-ethers (1) of phenols, thiophenol, and catechols have been prepared in good yield by their alkylation with (acyloxy)alkyl α-chlorides or iodides in acetone in the presence of K2CO3.In addition, one example of an (acylthio)alkyl α-ether was prepared.Either partial or complete acylation rather than alkylation on oxygen took place if the α-iodide was not used except for reactions of 3-chloro-1(3H)-isobenzofuranone with phenol or catechol or the reaction of (benzoylthio)methyl chloride with β-estradiol.It is suggested that more alkylation takes place with iodide as a leaving group because of the tighter transition state that develops with the better nucleofuge.The alkylation reaction sequence has two advantages for the synthesis of 1: (1) the mildness of the reaction conditions and (2) the wide variety of acyl and alkyl groups that can be incorporated into the product through the (acyloxy)alkyl α-halides.

The electrochemical reduction of some S,S-diarylbenzene-1,2-dicarbothioates (1) in aprotic medium leads to 3,3-bis(arylthio)phthalides (2) in almost quantitative yield.The reaction is an example of an electrochemically induced chemical reaction.SRN1, in which no external nucleophile is needed and for which the required amount of charge is less than 0.1 F mol-1.
