The invention provides a preparation method for a polysubstituted 2-aminooimidazole compound. The compound is described in a formula (III). The preparation method comprises the following steps: subjecting propargylamine as shown in a formula (I) and carbodiimide as shown in a formula (II) to cyclization reaction in an aprotic polar solvent under the action of an alkaline matter and a condition of 25 to 125 DEG C temperature, then carrying out TLC trace monitoring until the reaction is completed, and post-treating obtained reaction liquid so as to obtain the polysubstituted 2-aminooimidazole compound. The preparation method for the polysubstituted 2-aminooimidazole compound provided by the invention has the following advantages: reaction raw materials are cheap and easily available; a noble metal catalyst is not needed in reaction process; reaction conditions are mild; the method in the invention is green and environmentally friendly and has high atom economy.
