Reaction of benzyl (2,2-diphenyl-4,5-hexadienyl)carbamate (4) with a catalytic 1:1 mixture of Au[P(t-Bu)2(o-biphenyl)]CI (2) and AgOTf (5 mol %) in dioxane at 25 °C for 45 min led to isolation of benzyl 4,4-diphenyl-2-vinylpyrrolidine-1-carboxylate (5) in 95% yield. The Au(l)-catalyzed intramolecular hydroamination of N-allenyl carbamates tolerated substitution at the alkyl and allenyl carbon atoms and was effective for the formation of piperidine derivatives, γ-Hydroxy and δ-hydroxy allenes also underwent Au-catalyzed intramolecular hydroalkoxylation within minutes at room temperature to form the corresponding oxygen heterocycles in good yield with high exoselectivity. 2-Allenyl indoles underwent Au-catalyzed intramolecular hydroarylation within minutes at room temperature to form 4-vinyl tetrahydrocarbazoles in good yield. Au-catalyzed cyclization of N-allenyl carbamates, allenyl alcohols, and 2-allenyl indoles that possessed an axially chiral allenyl moiety occurred with transfer of chirality from the allenyl moiety to the newly formed stereogenic tetrahedral carbon atom.

Synthesis, biological evaluation, and structure-activity relationships of 3-acylindole-2-carboxylic acids as inhibitors of the cytosolic phospholipase A2

3-Acylindole-2-carboxylic acid derivatives were prepared and evaluated for their ability to inhibit the cytosolic phospholipase A2 of intact bovine platelets. To define the structural requirements for enzyme inhibition, the carboxylic acid group, the acyl residue, and the moiety in position 1 were systematically modified. Furthermore, different substituents were introduced into the phenyl part of the indole. Replacement of the carboxylic acid group in position 2 of the indole with an acetic or propionic acid substituent led to a decrease of inhibitory potency. Enzyme inhibition was optimal when the acyl residue in position 3 had a length of 12 or more carbons. Conformational restriction of the acyl residue did not influence activity. Introduction of alkyl chains at position 1 of the indole with 8 or more carbons resulted in a loss of activity. However, replacing the Ω-methyl group of such compounds with a carboxylic acid moiety was found to increase inhibitory potency significantly. Among the tested indole derivatives, 1-[2-(4- carboxyphenoxy)ethyl]-3-dodecanoylindole-2-carboxylic acid (29b) had the highest potency. With an IC50 of 0.5 μM it was about 20-fold more active than the standard cPLA2 inhibitor arachidonyl trifiuoromethyl ketane (IC50: 11μM).

A series of acetylenic and allenic derivatives of 2-(5-methoxyindolyl)methylamine has been synthesized.The new compounds were studied as inhibitors of the A and B forms of the mitochondrial monoamine oxidase (MAO) from bovine brain, using (14)C-tyramine as the substrate and clorgyline and l-deprenyl as references.All the studied compounds were MAO inhibitors.However, these compounds either did not show selectivity (compounds 3a-3d, 4c, 4e, 4m and 4o) or they were selective for MAO-A (compounds 4a, 4b, 4d, 4f-4l, 4n and 4p).Some of the compounds showed a similar inhibitory potency for MAO-A and lower for MAO-B than clorgyline and the higher selectivity for MAO-A was about 2.5 times that of clorgyline.Selectivity was shown only by acetylenic and allenic potent inhibitors, but no simple relationship between inhibitory potency and selectivity was found.
