The invention provides a synthetic method of fluticasone propionate impurity EP-ZI. The method comprises the following steps: firstly, chlorosulfide and ethanol are used as raw materials to carry outa reaction to generate a compound A; the compound A reacts with thioacetic acid to generate a compound B; and the compound B reacts with a compound C to generate the impurity EP-ZI. The synthesis method is simple in process route, convenient in operation, good in selectivity and relatively high in yield. The synthesized fluticasone propionate impurity EP-ZI can be used as a reference substance fordetection and research of fluticasone propionate, and is applied to quality control of fluticasone propionate and related preparations thereof, and purity of fluticasone propionate crude medicines orpreparations of the fluticasone propionate crude medicines is controlled.
