Manganese-Catalyzed Selective Hydrogenative Cross-Coupling of Nitriles and Amines to Form Secondary Imines

Manganese complexes with tridentate PNN ligands have been synthesized as catalysts for hydrogenative cross-coupling reaction of nitriles and amines to form secondary imines. This reaction afforded a variety of unsymmetrical secondary imines in good yields with excellent selectivity. Investigation of catalyst intermediates indicated that an amido manganese complex may be the active catalyst species for this reaction. (Figure presented.).

The cytotoxic activity of two series of platinum(II) complexes containing the polyfunctional imines R1-CHN-R2 [R1 = phenyl or ferrocenyl unit and R2 = (CH2) n-CH2-NMe2 where n = 1 or 2) (1 and 2) or C6H4-2-SMe (3)] acting as a bidentate (N,N′) (4-7) or terdentate [C(phenyl or ferrocenyl),N,N′]- (8-10) or [C(ferrocenyl),N,S]- ligand (11) in front of A549 lung, MDA-MB231 breast and HCT116 colon human adenocarcinoma cell lines is reported. The results reveal that most of the platinum(II) complexes are active against the three assayed lines and compounds 6, 7 and the platinacycles 10 and 11 exhibit a remarkable antiproliferative activity, even greater than cisplatin itself, in the cisplatin resistant HCT116 human cancer cell line. Electrophoretic DNA migration studies showed that most of them modify the DNA tertiary structure in a similar way as the reference cisplatin. Solution studies of a selection of the most relevant complexes have also been performed in order to test: (a) their stability in the aqueous biological medium and/or the formation of biologically active species and (b) their proclivity to react with 9-methylguanine (9-MeG), as a model nucleobase. Computational studies at DFT level have also been performed in order to explain the different solution behaviour of the complexes and their proclivity to react with the nucleobase.

Biomimetic aerobic oxidation of primary benzylic amines has been achieved by using a quinone catalyst. Excellent selectivity is observed for primary, unbranched benzylic amines relative to secondary/tertiary amines, branched benzylic amines, and aliphatic amines. The exquisite selectivity for benzylic amines enables oxidative self-sorting within dynamic mixtures of amines and imines to afford high yields of cross-coupled imine products.
Intramolecular aminoalkene hydroamination catalyzed by magnesium complexes containing multidentate phenoxyamine ligands

The magnesium complexes L2MgiPr (L2 = 4-tert-butyl-6-(triphenylsilyl)-2-[bis((3-(dimethylamino)propyl)amino)methyl] phenoxyl) and L3MgiPr (L3 = 4-tert-butyl-6-(triphenylsilyl)-2-[benzyl((3-(dimethylamino)propyl)amino)methyl] phenoxyl) supported by potentially tetradentate and tridentate triphenylsilyl-substituted phenoxyamine ligands have been prepared and fully characterized. The X-ray crystallographic analysis of L2Mg iPr confirmed a monomeric structure in which only one of the amine side arms is bound to the four-coordinate magnesium atom. The free and coordinated side arms in L2MgiPr undergo an exchange process at 25 °C in solution, while the phenoxydiamine complex L 3MgiPr, on the other hand, shows no sign of fluxionality. Both complexes, as well as L1MgiPr (L1 = 4,6-di-tert-butyl-2-[bis((3-(dimethylamino)propyl)amino)methyl]phenoxyl), were shown to be competent catalysts in the cyclization of aminoalkenes. L 2MgiPr exhibited the best catalytic activity, and both triphenylsilyl-substituted complexes display zero-order rate dependence on substrate concentration and first-order rate dependence on catalyst concentration, whereas the sterically less hindered complex L1Mg iPr exhibits second-order rate dependence on substrate concentration. No Schlenk-type ligand redistributions were observed, and the catalytically active magnesium species was stable after prolonged heating to 120 °C, according to an NMR spectroscopic study.

The reaction of compound cis-[PtCl2(dmso)2] with ligands RCHN(CH2)3NMe2 (1a-1h) in which R is a phenyl group containing substituents such as Cl, F, Me, NO2 and MeO produced compounds [PtCl2{(CH3)2N(CH 2)3NCHR}] (2), as mixtures of Z and E isomers. When treated with an equimolar amount of sodium acetate in refluxing methanol compounds 2 gave cyclometallated platinum compounds 3 containing a terdentate [C,N,N′] ligand. The obtained compounds were fully characterized including structure determinations for compounds 3a, 3b and 3e. The effects of the substituents in the regioselectivity of the cyclometallation reaction and the reactivity of 3a and 3b with mono and bidentate phosphines were also studied.

A new series of 2-(4- and 3-substituted phenyl)-3--1,3-thiazolidin-4-ones were synthesized, characterized, and evaluated for their ability to inhibit the contractions induced by histamine on guinea pig ileum.The measurement of pA2 values suggested that the reported compounds showed H1-antagonism.The more active compounds 5,9 and 13 exhibited activity close to that of mepyramine.
