Synthesis of 3- and 6-substituted steroidal heterocycles as potential anticancer agents

Coupling via C-N linkage between 3-bromo- or 3-mesyl steroids and various heterocycles, including pyrimidines and imidazole derivatives, was accomplished by adapting general methods used in nucleoside synthesis. PMR evidence indicated the formation of the isomeric 3-substituted steroid derivatives, and in certain instances the formation of 6-substituted 3,5-cyclosteroids. Assessment was made of different steroid bromination methods. Presumptive activity against L-1210 lymphoid leukemia in female mice was observed for one of the isomers of 3-uracil-substituted 5-pregnen-20-one.
